I interviewed with an investment banking firm over three weeks ago. I called once. Should I call them again?

They seemed to really like me. I called over a week ago and the hiring manager was on vacation. I've been calling HR, not the hiring manager.

Don't listen to the above comment. Call again. You have to be persistant and show them you want the job. Force them to think about you.
Usually, you're suppose to call a day or two after the time period that they told you for which you should expect a decision. For example, if they said that you can expect to hear from them in 2 weeks, you can call after that time. So if the IB told you to expect something in a week, call them once a week after that. Do not push them to make a decision, otherwise they will not consider you any longer. You also do not want to appear as if you don't care. Good luck with your job search.
