Negotiating salary with a non-profit?

I have read that people should negotiate salary offers, even for non profits. However, is this appropriate when the job advertisement states specifically that, for example, the job pays $25,500 with benefits? There doesn't appear to be any wiggle room.

Of course you can negotiate with a non profit but sometimes the budgets are struck and they can't raise the salary, or if it's a unionized position they may not be able to change it.

The first step is to ask if the rate they offer you is negotiable. If they say no it's up to you to take it or leave it. If you really feel it is underpaid you can always ask for an extra perk such as another week of holiday or flexible hours. It depends how much they want you. Always wait until they have officially given you the job offer before you ask for more money.
There is always wiggle room. How far to push it is up to you.
